LAG-3 (lymphocyte activation gene-3, CD223), a molecule belonging to immuno- globulin superfamily, has structural homology to CD4 [1].
LAG-3 binds MHC class II via its D1 domain at 60 nM Kd, several orders higher affinity than that of CD4 for MHC class II [2].
